Olink® Technology
Olink® assays are built on Proximity Extension Assay (PEA) technology, a highly specific, dual-antibody method for protein detection. In this approach, two matched antibodies bind to the same target protein, each carrying a unique DNA barcode. When both antibodies recognize the protein simultaneously, their DNA tags come into close proximity and hybridize, creating a template for DNA extension. This newly extended DNA molecule is then amplified, and the signal is quantified using qPCR or next-generation sequencing, enabling precise measurement of protein abundance.
The Olink Signature Q100 and associated assay platforms support high-multiplex protein analysis with key advantages, including:
- Low sample requirements, enabling analysis from minimal plasma or serum volumes
- High sensitivity and specificity for detecting low-abundance proteins
- Wide dynamic range, capturing subtle and large-scale biological changes
- Compatibility with diverse sample types, including plasma, serum, CSF, and other matrices
Olink offers multiple panel formats—from mid-plex Target panels (48- and 96-plex) to customizable Olink Flex and Olink Focus assays—allowing researchers to tailor protein quantification to their study’s scope and goals.
Olink® Service Frequently Asked Questions
What is Olink® Proteomics and how does it work?
Olink® Proteomics uses the Proximity Extension Assay (PEA) technology to quantify thousands of proteins simultaneously from minimal sample volumes. Each assay employs dual antibodies linked to DNA oligonucleotides that hybridize when bound to the same protein, enabling highly specific detection through next-generation sequencing (NGS) or qPCR readouts. This approach provides exceptional sensitivity, reproducibility, and scalability for biomarker discovery and validation.
How can Olink® services accelerate biomarker discovery?
Olink® Explore HT, Explore, and Reveal services enable high-throughput profiling of over 5,000 proteins across hundreds of samples in a single run. By integrating Discovery’s automated workflows and in-house biorepository, researchers can shorten project timelines, identify novel biomarkers faster, and transition seamlessly to validation using Olink® Target, Flex, and Focus platforms.
What are the differences between Olink® Explore and Olink® Target platforms?
The Olink® Explore and Reveal platforms support large-scale, discovery-phase proteomics, measuring thousands of proteins with broad coverage and minimal bias. Olink® Target, Flex, and Focus are used for the next phase — targeted validation — allowing researchers to quantify specific biomarkers with high precision and customizable assay panels. Together, they provide an end-to-end proteomics workflow from discovery to validation.
How does Discovery integrate AI and bioinformatics with Olink® data?
Discovery Life Sciences combines Olink® proteomic data with AI-driven bioinformatics to uncover patterns across large datasets. Machine learning models identify statistically significant biomarkers, classify patient subgroups, and predict biological pathways involved in disease mechanisms. This integrated approach enhances interpretability and translational value from high-throughput proteomic studies.
What sample types are compatible with Olink® assays?
Olink® assays are compatible with a wide range of sample matrices including plasma, serum, cerebrospinal fluid (CSF), urine, dried blood spots, conditioned media, and tissue biopsies. Discovery’s validated workflows ensure consistent performance across all matrices, enabling flexible study designs and longitudinal analysis.
How much sample is required for Olink® assays?
Only 25 µL of sample is required for a full suite of Olink® Explore proteins, and as little as 1 µL for selected Target or Reveal assays. Discovery’s automated handling minimizes waste while maintaining reproducibility across replicates.
What types of research can benefit from Olink® Proteomics?
Olink® Proteomics supports biomarker discovery and translational research in oncology, cardiology, neurology, immunology, and metabolic disease. Discovery’s certified Olink® services enable both exploratory and validation studies, supporting applications from early discovery to clinical biomarker verification.
